Abstract Two new methods for the isolation of membranes of Photosystem II (PSII) from the green algae Chlamydomonas Reinhardtii are presented in chapter two. The protein subcomplexes which were isolated by these methods were spectroscopically and biochemically characterized. The electon transfer reactions in the oxidizing side of PSII were studied by EPR spectroscopy. The detection of hydrogen kinetic isotope effects on this reactions implies coupling of the electron transfer with proton transfer reactions for the photosynthetic oxidation of water. In this thesis, a new model for this process is presented. According to the model, the electron flow in the oxidizing side of PSII is coupled with a proton flow originating from the substrate water. Experimental data shown in chapters three and four support this model. The radical which is generated by illumination of Ca2+ -depleted PSII membranes produces the EPR signal known as "S3 signal". In chapter five, the spectroscopic characterization of this signal and a study of the properties are dependent on the experimental conditions which are used and the disrepancies in the experimental data reported in previous studies are the results of this dependency. By this study it is proved that the origin of the "S3 signal" is the radical Yz which is magnetically with the manganese cluster at the S2 state with a dipole-dipole interaction.
