Abstract Mitochondria are considered to be essential organelles, responsible for a series of different procedures important for cell viability. One of the roles they are responsible for is the oxidative folding of protein molecules, a process performed primarily by the Mia40-Erv1 pathway. This well-described pathway is considered to be crucial for the oxidative folding of proteins that function in the mitochondrial intermembrane space. Recent studies in other sub-cellular compartments revealed that oxidative folding of proteins through a similar conserved pathway (PDI-Ero1) requires the presence of antioxidant proteins, such as PrxIV. The aim of this study was to elucidate the role of Gpx3, a well-known antioxidant protein recently found through proteomic analysis to reside in the mitochondrial intermembrane space, as part of an alternative redox regulation pathway.
