Abstract Glutamate dehydrogenase (GDH) is a key enzyme of the nitrogen assimilation pathway, with other possible functions too. In our effort, to identify the copy number of genes which encode for GDH in Vitis vinifera (L.), was performed blot analysis in genomic DNA from Vitis vinifera cv Sultanina. Two genes that encode for the two subunits of GDH were identified. Screening procedure in λ FIX II genomic library of Vitis vinifera cv Sultanina, with probe of full cDNA from gdha gene. Nine clones interacting with the probe were isolated. In order to characterize the molecular organization of the two genes of GDH, we cloned in pGEM vector intersected parts from each clone. We obtained the intersected parts with PCR reaction. From the sequencing and the alignment of the clones, we characterized the full length GDHA gene while GDHB is in preparation. GDHA gene consists of 9 exons and 8 introns. We also recognized, cis acting elements like TATA box in GDHA promoter region.
