Abstract Histone acetylation is a well studied mechanism that leads to transcriptional activation. Histone deacetylase inhibitors have been used for several purposes such as cell cycle inhibition. In this report, the role of trichostatin A (TSA) is studied in the transcriptional induction of MHC class II molecules in the absence of their master regulator, CIITA. The genes that respond to TSA belong to diverse functional groups. Two major groups of induced genes are the histones and the MHC class II genes. They are co-localized on chromosome 6p21.3 forming a region of high transcriptional activity upon inhibition of deacetylation. The molecular mechanisms and the events that take place on the promoter of DRA gene were studied. The response of the MHC class II promoter to TSA is cell-type specific and involves hyperacetylation and reduction of histone methylation in the vicinity of the promoter. For the first time it is shown that HDACs associate with enhanceosome factors providing it with an active role, beyond CIITA recruitment, in class II negative regulation.
