Abstract Tpl2/Cot is a MAP3 serine/threonine kinase that participates in the regulation of various kinase signaling pathways and transcription factors. This kinase is found to be implicated in miscellaneous pathological entities referring to inflammation and cancer. NPM is a phosphoprotein of ~ 37kDa that bears a complex primary structure and has the capacity to shuttle between the nucleus and the cytoplasm. NPM acts as a molecular chaperone and plays a significant role in ribosome synthesis. NPM can also regulate the genome stability and is essential for the modulation of protein cascades which get activated under stress conditions. Operating in a wild-type form, mutated or as a part of chimeras, NPM is implicated in many types of malignancies. The present study intends to establish a structural and/or functional interaction between the abovementioned proteins and to prove a possible correlation between immunological and tumorigenic mechanisms in a molecular level.
