Abstract This work constitutes a pilot study, whose main objective was to address the potential problems that arise during the transition from the experimental model of yeast to mammalian cells. Conduit for the transition is the human analogue of a protein that reportedly plays an important role in the biogenesis of mitochondria. This protein, called Erv1 (the human analogue is ALR (Augmenter of Liver Regeneration)) is one of the two main players of the Mia40 pathway. This pathway is located in the intermembrane space of mitochondria, where oxidative folding of proteins with specific cysteine motifs takes place. The first steps to consolidate the experimental approaches were performed in this study which controlled the characteristics of ERV1/ALR protein in mammalian cells.
