Abstract Lung cancer is the leading cause of morbidity and mortality among men and women worldwide. It is now more apparent than ever before, that genetic mouse models that closely recapitulate carcinogenesis are needed. CRISPR/Cas9, the most powerful genome editing tool, has come to change the way we are approaching genome editing forever. In the presence thesis we have discussed about some of the many applications of the technology. Using CRISPR/Cas9 it is possible to delete small regulatory elements such as transcription factor’s binding sites, to overexpress the transcription of specific loci, to introduce specific mutations and model oncogenic translocations in vitro and in vivo. Indeed, here we show that our editing successfully led to the modelling of well established oncogenic alterations related to Lung Cancer, very closely mimicking these of the human disease.
