Abstract We studied the interactions of regulatory factors that bind X box of the promoter of the Class II molecules of the major histocompatibility complex. To this end five important factors were cloned and purified. Using GST pulldown assays, we found that RFX5 interacts in vitro with RFXANK, RFXAP, CREB and the Class II Transactivator (CIITA) applying GST. CIITA interacts in vivo with RFX5 as it is shown by coimmunoprecipitation experiments and it also interacts in vitro with RFXAP. Interestingly, RFX also interacts with P/CAF. Thus, RFX5 and CIITA are the first players that allow the formation of enhaceosome by recruiting the other architectural proteins and finally the essential coactivators like CBP or P/CAF.
