Abstract Carbon nanofibers are considered promising materials in developing reliable biosensors due to their unique electrochemical properties, high surface area, mechanical and chemical stability. The surface of these materials contains functional groups, which can interact with biomolecules, leading to their stabilization. Oxidation of carbon nanofibers provides the possibility of introducing new functional groups on their surface, lending them even more attractive immobilization matrices for enzymes. An increase in the number of surface groups allows the stronger interaction with enzymes via adsorption. Additionally the introduction of specific type of groups like carboxylic groups allows the covalently immobilization of biomolecules. Therefore the conjugation of chemically modified carbon nanofibers with enzymes can lead to the development of biosensors with better characteristics like sensitivity and stability. In this study carbon nanofibers have been modified with concentrated H2SO4 / HNO3 solution increasing the number of total acidic groups and especially the number of carboxylic groups on their surface. The oxidized carbon nanofibers have been utilized for the development of glucose biosensor for the detection and monitoring of glucose. Glucoze oxidase was adsorbed and covalently immobilized via carbodiimide chemistry on the oxidized nanofibers. The electrochemical characteristics of both biosensors and their ability to immobilize enzyme molecules were compared. In both cases it has been shown that the oxidized nanofibers are significantly better matrices for the development of advanced biosensor systems.
