Abstract Secretion and translocation of virulence proteins by plant pathogenic occurs by means of type III secretion systems. Type III secretion systems are multicomponent (>20) protein complexes. The molecular mechanism of the secretory process and the structure of these systems are still unknown. Protein-protein interactions among proteins of the type III secretion system and protein AvrPphB from the plant pathogenic bacterium Pseudodomonas AvrPphB from the plant pathogenic bacterium Pseudomonas syringae pv. phaseolicola were examined in this study. The AvrPphB protein is an Avr protein. It causes the hypersensitive reaction when translocated into the cytoplasm of resistant plants having the respective R gene. Translocation requires a functional type III secretion system. Three protein-protein interactions were identified using the in vitro method Far Western. The nature of these interactions and functional implications are discussed.
