Abstract To understand gene function, it is helpful to know where, when, how much a gene is expressed and what other genes are coexpressed with it. By making possible the parallel measurement of gene expression of thousands of genes, microarrays appear as a tempting and valuable tool. Their potential for parallel expression changed the way biological questions were asked into a more global direction. DNA microarrays estimate the expression of each gene by measuring the concentration of the corresponding mRNA. So, the quality of the investigated mRNA is of prime importance for the microarray experiments. In a degraded RNA sample, cleaved mRNA molecules may no longer be representative of the absolute amount of transcripts and of their relative abundance. The conventional methods for RNA quality assessment are not considered to be sufficient. Therefore the necessity arises to develop sensitive methods for estimating the integrity of transcripts after RNA preparation. Here we propose a new quality control method for RNA by means of quantitative reverse transcription real-time PCR assaying the integrity of a long constitutively expressed and abundant transcript.
