Abstract This work is part of our project concerning human chromosome 10 genomic mapping. It has been based on a preliminary integrated genetic, cytogenetic and physical map constructed in our lab for this chromosome. The aim was to contribute to the construction of a detailed physical and expression map of 10q23-q25.1 chromosomal region. This region is under intensive investigation by many groups, because of its high gene-content and its correlation with diseases, including cancer, developmental abnormalities and neurological syndromes. For this purpose, a collection of approximately 500 YAC, PAC and BAC clones were isolated, based on their content in ESTs and genetic markers. Several groups of the above mentioned clones have been organised in high resolution contigs by a variety of approaches, including detailed fingerprinting by restriction enzymes. High resolution fingerprining contigs were constructed for some of the genomic clones, based mainly on BACs. Our ultimate aim is to build a sufficient composite (physical and transcriptional) map in a region of about 20Mb, representing most of the 10q23-q25 region. This will be the basis for the constuction of a sequence-ready tilling path to be used for the determination of gene organization therein. Furthermore, it will contribute to the analysis of interesting genes of the region and elucidation of their possible role in human diseases.
