Abstract Multiple sclerosis (MS) is an autoimmune neurodegenerative disease of central nervous system (CNS). Microglia are tissue – resident macrophages implicated in central nervous system development, homeostasis and response to injury. In the case of MS microglia are the first cells to arrive to the MS lesions to clear tissue debris in order to restore CNS homeostasis. However, microglia cells are proposed to also contribute to MS development. This hypothesis suggests that impaired microglial autophagy manage to affect MS development. Here, we demonstrate that microglial autophagy is induced upon experimental autoimmune encephalomyelitis (EAE) in mice, a well established mouse model to study MS. Primary neo – natal microglia treated for 18h with LPS, IFNγ or IFNα failed to recapitulate the induced autophagy upon EAE development. Surprisingly, LPS treatment manage to induce the p62 expression and decrease autophagy in primary microglia even though LPS treatment is well established to induce autophagy in macrophages through TLR4 signaling. Further research need to be done in vivo in order to unravel how and if microglia contribute to EAE/MS development.
