Abstract Autophagy has classically been described as a process that delivers cytoplasmic proteins and organelles to lysosomes for degradation. However, autophagosomes (AVs) have also been shown to fuse with the plasma membrane, thereby allowing the secretion of soluble proteins to the extracellular space and the targeting of transmembrane proteins to the plasma membrane. In concert with the notion that degradative and secretory AVs constitute distinct populations, confocal microscopy experiments demonstrated that Stx17 and Sec22b (the degradation- and secretion-specific AV SNAREs, respectively) have distinct patterns. Sequential centrifugation analyses also revealed Sec22b to be enriched in smaller cellular structures compared to Stx17, which could allude to its recruitment on the outer autophagosomal membrane early on in the autophagosomal biogenesis process. Genetic studies in mice have demonstrated the requirement of basal autophagic levels for neuronal function, while it is becoming apparent that autophagy also exerts functions in the pre- and post-synapse. The contribution of secretory autophagy, however, has not been examined as of yet. Preliminary data demonstrate that the outer membrane of murine forebrain-derived autophagosomes is rich in integral proteins. It is thus hypothesized that secretory autophagy could mediate neuron-specific targeting of plasma membrane proteins, and to that end, the process is also examined in non-neuronal cell types.
