Abstract Under normal physiological conditions electrons leaking from the electron transport chain, during aerobic respiration, reduce molecular oxygen, leading to the formation of Reactive Oxygen Species (ROS). Under stress conditions the production of ROS increases, while abiotic stress has been proven to induce enzymic mechanisms of ROS production as well. The cell possesses a range of enzymic or non enzymic mechanisms in order to maintain a stable concentration of ROS. One type of abiotic stress is salinity. Salinity is a complex stress factor incorporating osmotic and ionic stress as well as secondary stress factors such as oxidative stress. In this work an effort has been made to establish a connection between salinity stress and oxidative stress. Tobacco plants have been subjected to salinity stress of different intensity and duration. At the end of every treatment the ROS levels were determined and the special activity of the major antioxidant enzymes of the antioxidative mechanism was measured. The phenomenon was studied in planta by using whole plants and in in vitro with the use of leaf discs. In both systems intense stress led to the increased production of ROS and induction of the antioxidant enzymes. To assess the role of antioxidative defence towards achieving salinity tolerance genetically modified plants were used with modified enzymic activities. The study of the genetically modified plants led to the conclusion that increased activity if the antioxidative mechanism may lead to partial protection of the plant under salt stress conditions.
