Abstract Natural killer cell activity CNKal and MICC were analyzed in 25 untreated patients with myelodysplastic syndromes. using cytotoxicity assays and immuofluorescence with antibodies. We found that patients with MDS have normal number of periferal blood NK-cells and CD8+Cells. NK-cells from MDS patients express very low NKa which may be increased with recomdinant a-IFN and rh IL-2, although it remains bellow the lower limits of the control range. The cells exhibit normal tumor cell binding capacity, but fail to release sufficient amounts of NKCFS upon their interaction with NK-sensitive K562 cells targets or their stimulation with phytohainylytivin(PHA). MICC was also found sugnificantlly decreased. Our results syggest that the defective NKa in MDSpatients wight be due, at least parially to impaired release of NKCFS. This disturdance is rather the resylt of an intrinsic defalt of MDS NK cells in NKCFS production, than an impairment of activatory signal providedby the stimulatory K562 cell tragets. We postulate that the abnormally may represent an other festation of dysplastic hauuopoiesis.
