Abstract The Enhancer of split complex of Drosophila melanogaster constitutes a very well studied system of a gene cluster. The proteins that are encoded in that complex belong to two different groups, the basic-Helix-Loop-Helix-Orange and Bearded, and participate in the regulation and execution of Notch signaling. The bHLH-O proteins of the complex play a significant role in the early development of the nervous system, where through the procedure of lateral inhibition, they specify the number and the spatial organization of the cells of the nervous system. The complex is conserved in Tetraconata, consisted of four genes in Crustaceans, three of which are bHLH-O and thirteen in Drosophila melanogaster, in which due to duplication it consists of seven bHLH-O. In this paper, we study the phylogenetic relationships between the proteins encoded in the complex in Tetraconata as well as the functional conservation of E(spl) proteins of Apis mellifera and Daphnia pulex. The interactions between the E(spl) and the proneural proteins is conserved for all the Apis mellifera proteins and for the two out of the three of Daphnia pulex. Also, they bind in the same motif on the DNA and at last, they mimic phenotypes of E(spl) overexpression in transgenic lines of Drosophila melanogaster. Thus, we demonstrate that the gene regulatory network responsible for the neurodevelopmental processes are conserved both at the sequence level and at the level of protein interactions.
