Abstract The protein superfamily of DEDD exonucleases consists of several members, which fulfill variant and important functions in RNA metabolism in a large spectrum of organisms. From participating in pathways of tRNA and rRNA processing and maturation in bacteria, to participation in DNA polymerase complexes as proofreading subunits, maturation of rRNAs and in regulation of RNAi pathways, DEDD exonucleases have attracted strong scientific interest over the last years. This interest is focused on their action. Their preference for double stranded RNA molecules that contain single stranded 3’ ends as substrates seems to dictate their function. The interest around a subgroup of this family, the so called Eris, which participate in rRNA and siRNA pathways (even though assumptions have been made for their participation in other processes such as the maturation of 3’ end of histone mRNAs), boosted scientific effort towards the characterization of their members in an increasing number of organisms. This dissertation is a part of this effort and its main goal was to characterize the ΕRI member, AtERI‐LIKE1 from Arabidopsis thaliana, which is the first member ever found to be localized into the chloroplasts. In a series of in vitro experiments AtERI‐LIKE1 was tested against a number of possible substrates (ssRNA, dsRNAi‐like, 5 S rRNA, 5,8 S rRNA) to determine whether it could bind to or process them. The results did not definitively show interaction between ΑtERI‐LIKE1 and the tested substrates. However, western blotting with anti‐AtERI‐LIKE1 serum against protein extracts from plant tissues indicated that more than one isoform of ΑtERI‐LIKE1 may be expressed in Arabidopsis thaliana plants.
