Abstract This study describes the development of a novel methodology for the quantitative compositional analysis of P.D.O. (Protected Denomination of Origin) Cretan Graviera cheese by combining NMR spectroscopy and chemometrics for its authentication compared to other Greek yellow hard cheeses. A large number of yellow cheeses originating from various cheese making producers from the island of Crete were analyzed, as well as cheese samples produced in other parts of Greece. The hard yellow cheese samples studied included Graviera (Cretan and non Cretan), Kefalotyri cheese, Kefalograviera cheese, Graviera Light and Smoked Graviera, that have been aged for different amounts of time. Two separate experimental protocols using solvents of different polarity were used for the analysis of the graviera samples: water was used for the extraction of aminoacids, organic acids, alcohols and other low MW compounds, while chloroform was used to extract the lipid part of the graviera cheese (linoleic, linolenic and saturated acids, triglycerides, diglycerides, etc.) NMR spectroscopy was used to determine the chemical composition of the polar and non-polar graviera extracts and quantitatively measure the metabolic profile of both extracts. The metabolite profiles and the bucketed full Η NMR spectra of the graviera samples, were analyzed using suitable multivariate statistical analysis techniques to successfully develop metabolomic models that can differentiate the graviera samples according to their geographical origin (PDO Cretan or non-Cretan, Heraklion – Rethymnon), and determine which metabolites or spectral regions were contributing to this successful discrimination.
