Abstract The primary purpose of this study was to evaluate the potential to quantify the different neuronal populations in the retina of healthy Sprague–Dawley rats in an accurate quantitative way by using flow cytometry. Rats were killed and the eyes were enuclated to achieve retinal dissection and then dissociation by trypsin. The cells were mechanically dissociated into a single-cell suspension. The dissociated cells were permeabilized and stained with the primary antibody and afterwards with the secondary. Αt least 100.000 healthy cells were analyzed with a FACScalibur and FlowJo software. Dead cell and cell debris were exluded from analysis by gating with forward scatter (FCS, cell size) and side scatter (SSC, cell complexity) as indicators. The primary antibodies were anti-rhodopsin against photoreceptors, anti-Protein Kinace C against bipolar cells, anti-calbindin against horizontal cells and antimicrotubule associated protein 1 against ganglion cells. Quantification of the above populations was possible using flow cytometry. In this study, the photoreceptors had the 53.99%, the ganglion the 7%, the bipolars the 2%, the horizontal the 1.5% in the hole mixed retinal population. The experiments were repeated three times and these measurements are the mean value for each cell category. Flow cytometry can be used to quantify the different neuronal populations in control healthy eyes and this verification will be very useful in future in studies in apoptosis of these cells.
