Abstract Intestinal cholesterol absorption plays an important role in determining the levels of circulating cholesterol. Recently a new pharmacological agent, ezetimibe, was released that reduces cholesterol absorption and plasma cholesterol levels. The molecular target of ezetimibe is likely Niemann-Pick C1 Like 1 protein (NPC1L1), which is expressed predominantly in the small intestine . Inactivatιοn of NPC1L1 in mice results in a 70% reduction in the absorption of orally administered radiolabeled cholesterol, which is similar to the reduction seen in wild-type mice treated with ezetimibe. The molecular mechanism by which NPC1L1 transports cholesterol is unknown. Previously, a post-doctoral fellow in the Hobbs/Cohen laboratory observed a cholesterol-associated change in the migration of NPC1L1 to a higher apparent molecular mass on a native gel. The first aim of my project is to determine if this higher molecular weight band represents a dimer of NPC1L1. Initial experiments have been performed by expressing differentially-tagged versions of NPCC1L1 in Chinese Hamster Ovarian(CHO-K1)cells and determining if the two forms of NPC1L1 co-immunoprecipitate (Co-IP).
