Abstract The development of most types of neurons is under the control of a small family of growth factors called neurotrophins. The first neurotrophic factors discovered are NGF (nerve growth factor) and BDNF (brain-derived neurotrophic factor), which are secreted by the target tissue and have the ability to support the survival of neurons and neurites during the development. NGF binds to the TrkA receptor, BDNF and NT-4 to TrkB, and neurotrophin NT-3 interacts predominantly with TrkC receptors, and all members of the neurotrophin family bind with lower affinity to p75NTR. Recent studies have been associated with the attenuation of neurotrophin levels with neuronal dysfunction and degeneration observed in the aforementioned pathological conditions. Recently, the endogenous neurosteroid DHEA was found to be able to block neuronal apoptosis by binding to neurotrophin receptors. However, DHEA is metabolised in vivo to steroids and can affect the endocrine system through the development of neoplasms. To overcome this undesirable effect of DHEA, avoiding the adverse effects of its metabolism, synthetic molecules were developed depending on DHEA. The present study focuses on the characterization of 8 newly synthesized neurotrophin-like small molecules. By utilizing phosphorylation assays, we assessed and tested for their time-dependent ability to bind and activate the TrkA and TrkB receptors.
