Abstract The field of bacterium virulence is very important and is directly involved with protein biotechnology science. One of the most basic bacteria studied in this field is the opportunistic pathogen Pseudomonas aeruginosa, which is directly related to hospital diseases. Its ability to quickly develop resistance in a variety of antibiotics, in combination with the restricted antibiotic choices, makes the understanding of its virulence bases very important in order to design new drugs. The protein MvfR is a LysR type transcriptional regulator, which plays important role in the virulence of this pathogen, by controlling the expression of many virulence factors, as well as, part of the type VI secretion system. The aim of this work is to define the three dimensional structure of the MvfR, in order to study its interactions with other molecules. The first results from a small scale expression and purification indicated that the protein is soluble. Therefore large scale purification was done in order to perform the first crystallization screens. Finally, expression and purification in Se-methionine nutrient medium was done in order to perform the MAD method, to get the atomic model of the molecule. After that, big and well shaped crystals were produced, which are about to be used for data collection in synchrotron, in order to analyze them and get the atomic model of the protein.
