Abstract In parallel worn on lab, we have shown accumulation of ssDNA and dsDNA in different cell types (pancreatic and microglia cells) that lack ERCC1. Accumulation of cytoplasmic DNA has been shown in both cases to be connected with acute interferon-related innate immunity responses. In the case of microglia cytoplasmic DNA driven immune responses were shown to lead to a strong neurodegenerative phenotype. Exosomes are secreted vesicles and part of the normal intracellular communication. They have been implicated in numerous physiological and pathological states of cells and recently proposed as potential specific cargo/drug delivery system. We propose the production of engineered exosomes that will be targeted to specific cell types and will carry a nuclease able to degrade the cytoplasmic DNA thus reversing the acute inflammatory phenotype.
