Proliferation of toxin producing microphytoplankton in marine waters can lead to the formation
of Harmful Algal Blooms (HABs) with environmental, economic, social and public health
impact. Amongst the most prominent toxin producers are members of the Alexandrium genus,
who are known to produce Saxitoxins (STXs). Surveillance agencies monitor saxitoxin in
seafood and Alexandrium abundance in waters with effective, yet time-consuming methods.
This highlights the need for faster methodologies that enable quicker responses.
The focus of this Master’s thesis was to develop an assay for Alexandrium STX monitoring
that is compatible with on-site testing, at concentrations relevant to statutory monitoring
requirements. Recombinase Polymerase Amplification (RPA), is a rapid and portable
alternative to PCR, that meets the requirements for field-based devices. With that in mind, an
RPA assay was designed to target the locus of the fourth domain of the first enzyme (sxtA4)
that is involved in saxitoxin biosynthesis.
The first iterations of the assay sought to target a single species, Alexandrium minutum. After
several modifications of the primers and the incorporation of chemical additives, adequate
specificity results could not be achieved. Therefore, a “universal” sxtA4 assay, that targeted all
Alexandrium species, was developed and optimised. The assay demonstrated inclusivity to all
tested species (n = 4). The limit of detection of the assay was 103 sxtA4 copies for DNA and
104 copies for RNA. This optimized assay provides a novel tool with potential applications in
routine HAB monitoring.
