Abstract The Notch transmembrane receptors are activated by transmembrane ligands of the DSL family, which are subdivided into the Delta and Serrate / Jagged subfamilies in higher metazoans. Activation takes place exclusively in trans configuration (ligand and receptor in adjacent cells), whereas cis interaction downregulates Notch Signalling. Even when Notch and DSL are engaged in trans, a necessary prerequisite for active signal transduction, is the activation of DSL proteins by a ubiquitin E3 ligase. Work from our lab and others over the past decade has characterized two families of RING domain E3 ligases which have the ability to activate DSL proteins, Neuralized and Mindbomb1 by ubiquitylation. Ubiquitylation of plasma membrane proteins is a signal for endocytosis, as well as for further sorting steps in intracellular trafficking. Indeed coexpression of these ligases with Delta or Serrate causes their recolocalization from plasma membrane, to endosomes which are not yet well characterized. Here we have used Delta wild type protein and several endosomal markers to observe its intracellular trafficking and distribution in I) endogenous levels of expression and in II) controlled overexpression. We show that fast recycling endosomes (Rab4+vesicles) collocalize almost perfectly with Delta positive vesicles. On the contrary early (Rab5 positive), and late (Rab7 positive) endosomes have lower scores of colocalization.
