Abstract The objective of the present work is the development and optimazation of an analytical method for carbohydrates. The developed method includes the chromatographic separation of carbohydrates their derivatization in a Post Column Derivatization (PCD) system and finally the fluorometric detection of the derivative. The use of PCD instrument with two thermostated reaction coils makes the on-line reactions possible. Initially, the periodate oxidation of carbohydrates takes place with the use of 0,05 M sodium periodate at 120 0C. Subsequently the derivatization reaction of the formaldehyde formed from the above reaction is carried out with a 6 mM 4-amino-3-penten-2-one solution containing also 0,5 M sodium acetate and 0,2 M sodium thiosulfate. The detection is based on the fluorescence of the derivative formed which is monitored at 520 nm (emission) with an excitation wavelength of 402 nm. The detection limits of the method are 0,42 μg for glucose, 0,3 μg for mannose and 0,22 μg for fructose.
