Abstract Several cytoskeletal changes occur during cell transformation while alterations in cytoskeletal protein organization and morphology have been reported in neoplastic cells. Based on these reports severan agents, which intersact with cytoskeletal proteins have been considered as cancer chemotherapeutics. The aim of the study was to estimate the tubulin and actin polymerization dynamics as well as the cytotoxic action of triethyllead and cytochalasin B in normal and leukenic lymphocytes. We studied the expression levels of total tubulin amounts and the radio of polymerized tubulin in normal lymphocytic leykemia by quantitative immunoblot analysis. The alterations in the actin polymerization dynamics was studied by DNase I inhibition assay and fluorimertric measurements. Finally the time and dose-dependent effects of triethyllead and cytochalasin B in normal and maliguant lymphocytes was estimated in primary cell culture experiments. The result indicate that all tested leukemic cells are characterized by decreased polymerization state of actin and tubulin which reflects increased instability of microfilaments and microtubules respectively and increased suscepribility to specific anticytoskeletal agents such as triethyllead and cytochalasin B that could be further investigated as potential anticancer agents.
