Abstract This thesis includes research results concerning the design and synthesis of a series of fluorescent probes for intracellular cations. Retrosynthetic approaches are proposed, synthetic pathways are described, and the fluorescence profile of the synthesized probes is presented. The thesis consists of four major thematic units. Fluorescent calcium probes, based on the known tetracarboxylate chelator BAPTA. Near-membrane calcium fluorescent probes, similar to the aforementioned probes, incorporating in their heterocyclic framework lipophilic chains that may function as anchors by association with the inner membrane surface. Fluorescent caged-calcium probes that can function simultaneously as both, caged-calcium and calcium probes, thus avoiding the need for a fluorescent calcium probe to be used when releasing ions from a caged-calcium system. Fluorescent zinc probes that combine the appropriate fluorophores with chelator moieties necessary for the detection of [Zn2+]. A detailed fluorescence profile study of the probes synthesized and an evaluation of their overall properties as cation sensors is presented at the end of each section.
