Title Insights into the role of nuclear hormone receptor NHR-85 in Caenorhabditis elegans ageing

Abstract Ageing is defined as the progressive accumulation of damage over time, leading to the disruption of physiological functions in most living systems. A major aspect of ageing is associated with energy metabolism, and this relationship has been extensively investigated. Ageing is known to be accompanied by a general decrease in energy levels and basal metabolic rate, as well as alterations in mitochondrial function. Despite reported findings, though, the association between ageing and energy metabolism remains largely obscure. In mammals, energy metabolism is regulated by a plethora of transcription factors, including peroxisome-proliferator-activated receptors (PPARs), which comprise a major family of nuclear hormone receptors. Of these, the well-studied PPARα and PPARγ, which participate in key procedures of lipid metabolism, such as lipogenesis, have been suggested to regulate inflammation and ageing. In this thesis we aimed to study a putative orthologue of the least studied isoform PPARδ, named NHR-85, in the nematode model Caenorhabditis elegans. Our results suggest a direct role for NHR-85 in lifespan regulation, which is associated with dynamics of neutral lipid content. Elucidation of the role of NHR-85 in this powerful in vivo model will contribute to our understanding of the conserved mechanisms of ageing and will direct the research on its putative mammalian counterpart towards the development of novel anti-ageing interventions.
