Abstract Many reports demonstrate the presence of s-MHC-I proteins at significantly high levels in the serum of patients suffering from various autoimmune diseases, while remaining at low levels in healthy individuals. Although some studies have been carried out to verify their action, their role in the course of the immune response remains unclear. It is known that CD8 T cytotoxic cells express a T cell receptor that recognizes a specific antigen as a complex with a class I molecule. In fact, CD8 specifically interacts with conserved aminoacids of class I molecules. Thus, class I except from competing with CD8, it can block TCR function. Therefore, major potential functional targets of soluble class I could be CD8 as well as TCR. Upon isolation of soluble class I molecules from serum of healthy mice, in the present work we study the effect of this protein on in vitro cultures of spleen cells as well as CD8+ T cytotoxic cells in the presence of an antigenic stimulus. The results point to a suppressive activity of soluble class I molecules, which concentrates to the cellular rather than the humoral type of immunity.
