Title AvrPphB, AvrPphF: Μελέτες δύο μολυσματικών παραγόντων από την Pseudomonas syringae pv. phaseolicola

Abstract Gram negative bacteria use a highly sophisticated secretion system for protein delivery into the eukaryotic host cells, the type III secretion system. The strategy they use is conserved among plant and animal pathogens. In this thesis the studies of the type III effectors AvrPphB and AvrPphF from Pseudomonas syringae pv. phaseolicola are demonstrated. The proteins were cloned to expression vectors carrying a 6-His tag and purification experiments took place. The two sitedirected mutants of AvrPphB G63A and G63E show proteolytic cleavage like the wild type protein. In the case of AvrPphF, the two open reading frames that constitute its operon suggest the common strategy of animal pathogens of a type III chaperone and its cognate effector. In silico studies predict a type III chaperon like secondary structure for ORF1 and produce a homology model based on CesT template (type III chaperone from EPEC E.coli). Co- purification experiments also suggest interaction between ORF1 and the aminoterminal part of ORF2.
