Abstract Virp1 (viroid binding protein 1), is a tomato protein that specifically interacts with viroids (A.E. Martinez, 2000). In order to elucidate the role of the protein in the cell and in the connection with viroid infection, we have designed and constructed a chimeric protein of Virp1 with GFP (green fluorescent protein). the vector pAVA321 with the gfp gene that was used, belongs to a series of vectors, especially designed for the construction of fusion proteins (A.G. von Arnim et al.,1998). The chimeric gene was transferred to the binary vector pART27, suitable for Agrobacterium mediated plant transformation. Stable transformation of N.benthamiana plants was achieved by cocultivation of leaf disks from N.benthamiana with Acrobacterium carrying the chimeric construction and further by using a regeneration protocol. This procedure resulted in the generation of N.benthamiana transgenic plants, which were analyzed by means of microscopic investigation, for localization of the chimeric protein.
