Abstract Systematic screening of normalized cDNA libraries is important for the determination of rare transcripts. A normalized library from K562 erythroleukemic cells was used for cDNA identification present in a cellular environment suitable for differentiation studies. Thirty-one out of 57 ESTs exhibited no significant matches to known entries. Among those we identified an unknown EST, HSIMBB246 (Genbank accession number: X93860), containing zinc finger motifs. In this study we report structure and expression data concerning the EST-derived novel human zinc-finger gene ZNF232 (Genbank accession number: Y15067) encoding for a polypeptide, member of the recently described SCAN/LeR box domain gene sub-family. The sequence of 1.5Kb cDNA predicted a 417 aa long ORF containing five Cys2His2 zinc fingers and a SCAN/LeR box. ZNF246, a single copy gene, maps to 17p12, in a region where other zinc-finger genes have been localized. ZNF232 coding region is organized in three exons potentially subjected to alternative splicing. Among several tissues, predominant expression was detected in skeletal muscle and heart; a major 7.0 Kb transcript is detected as well as several minor ones Elevated abundance was observed in embryonic carcinoma cells upon differentiation with chemical inducers. Minor transcripts exhibiting considerable size heterogeneity (2.0-2.5 Kb), were detected in various human and cell lines examined. The functional potential of the zinc-fingers and of the SCAN/LeR domain as well as the eleveated expression in skeletal/cardiac muscle is discussed; moreover, data concerning the genomic enviroment of ΖΝF232 at 17p12-13 are also presented.
