Abstract |
The aim of this work was the isolation and characterizaton of serine proteases as well as the study of midgut-specific promotes of the mosquito Anopheles Gambiae. The mosquitos of the genus Anopheles are the moste powerful vectors of malaria parasites.It is known that in the mosquito midgut, crucial parts of the malaria life cycle take place. Furthrmore the development of the parasite in the midgut depends on some mosquito serine proteases. The characterization of serine proteases and midgut-specific promoters could be potentially useful in the construction of transgenic mosquitoes that would be refractory to the parasite. This could help the control of malaria in tropical regions. As a first step, using the PCR technology, three genes were cloned(SP6A,SP6T and SP8T)that code for serine proteases. The sequence of these genes as well as the fact that they are transcriptionally active in early developmental stages, indicated that most probably they are involved in mosquito development. Furhtermore the promoters of two other serine proteases genes-ANTRYP1 and ANTRYP2 were analyzed using the P-element mediated transformation system of Drosophila Melanogaster. Transformation experiments using ANTRYP1 and ANTRYP2 genes along with their flanking sequences showed that the expression of these genes in the transformed Drosophila lines was restricted to the gut.(Abstract truncated)
