Abstract |
The present study describes the cloning and functional analysis of the human neural cell adhesion molecule TAX-1, as the chromosomal organization and regulation of the TAX-1 gene. Screening of a human cerebellum CDNA library, led to the isolation of the TAX-1 CDNA molecule, that showed 91% and 75% similarity at the amino acid level with its homologues TAG-1/F3 subgroup of the IG superfamily of neural cell adhesion molecules, that is six(6) immunoglobulin like domains of the C2 type(IGC2) which are followed by four(4) fibronectin type III repeats(FNIII) and a C-terminal hydrophobic region, characteristic to proteins that are linked to the cell membrane via a GPI linkage. The second IGC2-type domain presents the highest similarity among the three homologous proteins, whereas only the two mammalian preteins(TAG-1 and TAX-1)share in the second FNIII-type repeat the RGD tripeptide, a motif known to mediate recognition of fibronection by integrins. Stable expression of the TAX-1 molecule in the non-adherent cell line of D.Melanogaster Schneider-2(S2), showed a dramatic increase of cell adhesion in the transfected S2 cells. In analogous experiments using deletion constructs expressing either the IGC2-type domains or the FNIII-type repeats of the TAX-1 protein, we have shown that the FNIII repeats alone, are both sufficient and necessary for mediating the homophilic adhesive properties of TAX-1 protein(Abstract truncated).
