Alternative Title Structure and functions of the GCN5 protein of Scerevisiae. The route of the protein in the mediation of transcription

Abstract GCN5 mutant strains of S.Cerevisiae exhibit poor growth under conditions of amino acid limitation. We have established that GCN5 required for the proper function of the transcriptional activator GCN4. GCN5 also required for the activity of HAP2-HAP3-HAP4 tranxscriptional activation complex, GAL4 and GAL6 activators. Thus GCN5 is a new member of the recently revealed general class of transcriptional mediators(co-factors)that collaborate with certain specific DNA binding activators to promote high levels of transcription. We have cloned and sequenced the GCN5 gene. The deduced GCN5 protein conteins a regiion called bromodomain, which is couserved in other memebers of this new class of mediators. An other protein ADA2 is also required as GCN5. Genetic data suggest that the two protein interact and define one complex that mediates transcriptional activation.
