Abstract Protoplasts from most of the important agricultural species exhibit recalcitrance towards standard culture conditions.Oxidative stress may be implicated in the recalcitrance of plant mesophyll protoplasts. In this work, de novo protein synthesis and the antioxidative enzymes sod, cat and per were studied during the enzymatic isolation and the culture of therecalcitrant grapevine (Vitis vinifera l. cv sultanina) and the regenerating tobacco (nicotiana tabacum l. cv xanthi) mesophyll protoplats. Antixidative enzymes increased significantly in tobacco protoplasts, but remained low in grapevine protoplasts. It is suggested that oxidative stress, triggered during maceration, is implicated in the dividing potential of plant protoplasts.
