Abstract Tomographic modalities have been widely employed for the imaging of biological structures in order to study the physiology and mechanisms of living organisms. One very prominent technique that can reify the aforementioned goal is Optical Diffraction Tomography (ODT). ODT is an interferometric technique that maps the three-dimensional refractive index distribution of the specimen under study. It can provide quantitative information and accurate reconstructed images while taking the effects of diffraction into account. In this thesis, we extensively explore the backpropagation algorithm, through the lens of Born and Rytov approximations, for the two-dimensional and three-dimensional case of ODT. Additionally, we provide comparisons of the accuracy of the aforementioned approximations for various specimens. Furthermore, this research presents an application of ODT in the field of twophoton polymerization systems, proposing ODT as an advanced inspection tool for the interior of two-photon polymerized structures. Lastly, a simulation microscope pipeline is introduced, where we utilize a combination of different light propagation models to identify a computationally efficient approach to simulate light propagation through an experimental microscope setup.
