Abstract Plastics are today a huge problem for mankind, as they are particularly harmful to the environment in the long term and their management is not easy. A more effective solution than recycling is the replacement of conventional plastics with bioplastics, which are environmentally friendly, mainly due to their production from microorganisms and their biodegradability. A class of chemical compounds that have the required characteristics to replace conventional plastics are the polyhydroxyalkanoates (PHAs). The most widely studied PHA is polyhydroxybutyric acid (PHB). The bacterium Pseudomonas sp. phDV1 is a Gram-negative bacterium and is one of many microorganisms, which under stress conditions have the ability to produce PHB. In addition, this specific bacterium is capable of degrading aromatic waste, which is toxic to the human body. The PHBs in Pseudomonas sp. phDV1 are produced intracellularly and are organized into granules, called carbonosomes. These structures display on their surface a set of proteins, which is responsible for the structure (phasins), regulation (regulatory factors, PhaR), production (synthases, PhaC), and degradation (depolymerases, PhaZ) of PHBs. To optimize the production of PHB two mutants have been generated from Pseudomonas sp. phDV1, ΔphaR, and ΔphaZ, where the phaR and phaZ genes, respectively, have been deleted. This thesis focuses on determining the production of PHB in the WT, ΔphaR and ΔphaZ strains of the bacterium Pseudomonas sp. phDV1, under conditions of nitrogen deficiency, but also in the presence of phenol, in order to evaluate the efficiency of the mutants. Also, after the characterization of the polymer, the structure of PHBs is studied, with an emphasis on the proteins and interactions that exist on the surface of these granules. The structural study is done using cryo-EM, where data from electron microscopy are combined with proteomic analysis.
