Abstract The species Inula helenium belongs to the genus Inula (Asteraceae) and is being used in traditional medicine until today, mainly for the cure of respiratory problems and skin diseases, due to its antibacterial and anti-inflammatory properties. According to studies, the strong bioactivity of the plant is attributed to the eudesmanolides which contains, and mainly to the alantolactone (1) and isoalantolactone (6). That was the main incentive for the isolation of the compounds, which were obtained in high purity after extractions of the plant’s dried roots and application two separate isolation techniques. The isolation took place either after multiple column chromatography separations or after repeated recrystallization. During this procedure, two more secondary metabolites from the eudesmanolide class were isolated and they were found structurally similar to the aforementioned compounds, namely 11,13-dihydroalantolactone (11) and 11,13-dihydro-isoalantolactone (12). Besides the isolation of the natural components, the modification of their structure was considered as necessary for the structure-activity relationship studies and biological tests. Thus, each compound was derivatized as its nitrile, the corresponding acid, as well as its azide derivative and the corresponding amine. The secondary metabolites and their derivatives were characterized via NMR spectroscopy, GC-MS and HRMS. Lastly, antioxidant studies were conducted on compounds 1, 6, their derivatives and on the methanolic and hexane plant extracts, through the method of the DPPH radical. The study revealed a strong antioxidant capacity of the methanolic extract and a moderate antioxidant activity of alantolactone amine 5a. Similar studies are underway in our laboratory.
