Abstract The focus of this thesis was on the study of cytochrome ba3 oxidase from Thermus thermophilus and on the drug of bleomycin (BLM). Cytochrome ba3 oxidase catalyzes the reduction of O2 to H2O and that of NO to N2O. The active center consists of a heme a3-CuB binuclear center and as a prosthetic group there are a low spin heme b and a homodinuclear CuA complex. We used as main spectroscopic technique the time-resolved step-scan FTIR, a structure-sensitive tool, and through the model complex of cytochrome ba3-CO, we detected and characterized important properties of the binuclear center, observed for the first time in analogous systems. In the second part of this thesis we studied the CO-Fe-BLM complex in the absence and in the presence of DNA. The FTIR spectra revealed no changes in the C-O vibrational mode indicating the absence of interactions between the BLM and the DNA in the active center of the complex.
