Title Mechanisms of local translation in the adult nervous system after injury

Abstract Neurons of the central nervous system (CNS) are characterized by reduced regeneration capacity compared to peripheral nervous system (PNS) neurons. A determinant of this difference, besides the environmental factors, is the intrinsic ability of injured axons to initiate a cell regeneration program. Local translation is a “key” process during injury response, as the majority of injury signals that are retrogradely transported to the cell body in order to modify the gene expression programs, are locally synthesized at the injury site. My thesis investigates the intricate signaling pathways associated with local translation during injury of adult CNS axons, and how the presence of Mena, a protein essential for both cytoskeleton dynamics and local translation, affects this response. Our data indicate that proteins with crucial roles during injury response such as mTOR, importin beta 1 and ERK ½, both in the presence and absence of Mena, are downregulated in injured CNS axons, in contrast to PNS where their upregulation is well-established by previous studies. These findings suggest that CNS likely fails to initiate a successful injury response, and further supports the notion that the limited capacity of injured CNS axons to locally translate/activate proteins implicated in pro-regenerative signaling pathways, significantly contributes to their reduced intrinsic ability to regenerate.
