Title Probiotic effect on inflammation and infection

Abstract Ulcerative Colitis is an inflammatory disorder characterized by significant gut microbial alterations. In this context, probiotics emerge as a promising avenue for modulating the microbiome and addressing this chronic, relapsing inflammatory disorder which lacks effective treatment options. This study investigates the anti-inflammatory effect of Pediococcus acidilactici (SK2), isolated from the human gastrointestinal tract, in a DSS-induced colitis mouse model. To this aim, mice were supplemented with SK2 and subjected to DSS administration. Real-Time qPCR and ELISA confirmed the anti-inflammatory effect of SK2, indicated by reduced levels of pro-inflammatory cytokines and chemokines. Additionally, qPCR demonstrated enhanced expression of mucus and tight junction genes, suggesting the beneficial role of SK2 on epithelial barrier integrity. 16S rRNA sequencing revealed minor alterations in gut microbial composition. Further preclinical and clinical studies are necessary to validate the safety and efficacy of P. acidilactici, potentially advancing its use in prevention and treatment of ulcerative colitis.
