Abstract One of the main brain regions that controls memory and is affected in the early stages of Alzheimer's disease (AD) is the hippocampus with its dentate gyrus (DG) subregion being involved in the input information flow from other brain regions and combines newborn and pre-existing neurons, astrocytes and microglia. Chronic stress is considered a major risk factor for AD through the deleterious effects of elevated levels of glucocorticoids (GC) that activate cellular mechanisms of AD pathology e.g. hyperphosphorylation and toxic aggregation of Tau protein. However, the effect of chronic stress on DG in AD pathology remains unclear. Thus, these Master studies aim to understand the effect of chronic stress and the interaction between different types of newborn and pre-existing cells of DG in Tau pathology. Our findings show the essential contribution of neuronal, but not microglial, glucocorticoid receptors in the stress-induced reduction of neurogenesis and astrogliogenesis in DG in Tau brain pathology. These findings provide new information on the detrimental effect of chronic stress on the complex neuron-microglia interaction that impairs DG plasticity in Tau pathology
