Abstract Our goal was to elucidate the IFN-a induced alterations on the degradation process of c-myc mRNA.Intermediate degradation products were detected in trasfected cells overexpressing structurally identical to normal c-myc mRNA.It was demonstrated that c-myc mRNA degradation follows two pathways.Rapid deadenylation results in the accumulation of oligoadenylated c-myc mRNA due to a endonucleolytic cleavage without prior deadenylation.Exposure to INF-a enhances this mechanism. Inhibition of protein synthesis does not prevent the formation of products of endonucleolytic cleavage.The traslability of the oligoadenylated c-myc mRNA generated in vivo,in the presence of cordycepin (3'dA) was determined.Our data indicate that oligoadenylated c-myc mRNA synthesized in the presence of cordicepin is more efficient as a template for protein synthesis compared to its counterart with heterogeneous distribution of poly(A) tails c-myc mRNA of control cells. The data suggest that the naturally generated oligoadenyl c-nyc mRNA through the deadadenylation process possibly represents the translationally active form of c-myc mRNA.
