Alternative Title Study of the activity of glucolytic enzymes during cell transformation with the T24-H-RAS oncogene

Abstract The aim of the study was to eludicate the role of the H-RAS ongogene in the regulation of glycolytic enzymes, since glycolysis is thought to be the main energy source of tumor cells. We develooped for this reason rat fibroblast and epithelial cell lines in which transcription of the exogenous human T24-H-RAS gene is regulated by addition of IPTG in the culture medium. We found that overexpression of H-RAS leads to activation of phosphoglycerate kinase, enolase and pyruvate kinase in fobroblasts but not in epithelial cells. This activation, however, did not result in alteration of the total glycolytic rate. We propose that these activations concern non glycolytic function of the above enzymes, as have been recently described.
