Abstract |
This study investigates the optical response of a novel anisotropic medium and its advantages in Light Sheet
Fluorescent Microscopy (LSFM). LSFM, renowned for its high resolution and multi-dimensional imaging,
is based on a single plane illumination of the sample using a laser beam shaped into a light sheet. This
investigation aims to provide deeper insights into the properties of this newly developed medium and its
potential applications in enhancing LSFM imaging quality. The study encompasses an exploration of light
propagation in a medium, providing the theoretical background in scattering, and the Spatial Light Modulator
operation in wavefront shaping. Experimentally, the research involves fabricating the anisotropic scattering
medium, the goniophotometer analysis of its angular intensity. Additionally, an analysis of its surface
morphology from SEM images was performed, as well as a reproducibility evaluation of the fabrication
process. Results demonstrate a high correlation in the medium’s behavior with a typical diffuser, due to the
dispersed energy distribution across one direction. The SEM analysis highlights the statistical properties of
its surface which can be used in future optical simulations and the reproducibility assessment showcases a
general trend towards similar values of the light sheet.
