Abstract |
The ability of many plant pathogenic bacteria to elicit the hypersensitive response (HR) on resistant host plants or on non-host plants, as well as to be pathogenic in their host plants is dependent on the presence of a cluster of genes, called hrp (hypersensitive reaction and pathogenicity). Certain hrp genes encode components of a specialized protein secretion pathway, called "type III" secretion system that appears to be dedicated to the secretion of virulence proteins in both plant and animal pathogens. Nine of the hrp genes have homologs in animal-pathogenic Yersinia, Shigella, and Salmonella spp., and these have been renamed hrc (for HR and conserved). In order to examine the biological activity of a truncated version of AvrPphB protein (AvrPphB-62G63M), lacking the first 62 amino acid residues, we used a transient expression system, based on virus PVX202 (Potato Virus X). The expression of AvrPphB-62G63M protein in Nicotiana benthamiana plants caused a severe necrosis, suggesting that the elimination of the 62 amino acid residues, had little affect on the ability of AvrPphB protein to trigger HR. In addition, hrp genes, belonging to the hrpC transcriprtional unit, were cloned out of the same pathogen. In order to understand the structure and the functional role of the type III secretion apparatus in plant pathogenic bacteria, we investigated the possible interactions among the components of the Hrp pathway.
