Title Analysis of the contribution of chromatin regulators in the process of adipogenesis

Abstract Obesity is a metabolic disorder characterized by abnormally increased fat accumulation within adipose tissue. Increased prevalence of obesity has rendered it a major global public health issue. In an effort to elucidate the mechanisms governing obesity, research studies have focused on adipogenesis, a major process involved in the adipose tissue formation and function. To this end, transcriptional regulation of adipogenesis has been extensively studied, while epigenetic regula-tion of adipocyte differentiation has only recently attracted attention. In this study, we examined the role of two members of KDM7 family of histone demethylases, PHF2 and PHF8. In contrast to previous reports, knock down of PHF2 facilitates adipogenesis while overexpression seems to hinder it. In addition, PHF8 also seems to have a negative role during adipocyte differentiation, as 3T3-L1 cells with knock down of PHF8 display increased lipid accumulation and increased expression of adipogenic genes. Taken together, these data suggest that PHF2 and PHF8 are neg-ative regulators of adipogenesis. Future studies confirming and expanding the findings reported here are required in order to conclusively identify the role of PHF2 and PHF8 during adipogenesis.
